NU7441 (KU-57788) verhoogt de persistentie van γH2AX-foci na door ioniserende straling geïnduceerde DNA-schade. Het (0,5 μM of 1 μM) verhoogt aanzienlijk de G2-M-accumulatie geïnduceerd door ioniserende straling en doxorubicine in zowel SW620- als LoVo-cellen. Deze verbinding veroorzaakt persistentie van door doxorubicine en ioniserende straling geïnduceerde dubbelstrengs-DNA-breuken en vermindert ook enigszins de homologe recombinatieactiviteit in DNA-PK-proficiënte M059-Fus-1 en DNA-PK-deficiënte M059 J menselijke tumorcellen. Het remt UV-geïnduceerde RPA p34 hyperfosforylatie op een dosisafhankelijke manier in zowel cellen die polymerase η missen als cellen die het tot expressie brengen. NU7441 verhoogt de niveaus van -geïnduceerde γH2AX-foci en vermindert dienovereenkomstig de -geïnduceerde celdood in chronische lymfatische leukemiecellen. Het remt ook -geïnduceerde DNA-PKcs-autofosforylatie en -herstel in chronische lymfatische leukemiecellen. Deze verbinding vermindert de frequentie van NHEJ en verhoogt de snelheid van HDR na Cas9-gemedieerde DNA-splitsing. |
