Flavopiridol werd aanvankelijk gevonden om de epidermale groeifactorreceptor en proteïnekinase A te remmen (IC50 = 21 en 122 μM). Later werd aangetoond dat Flavopiridol celproliferatie remt bij fysiologisch relevantere concentraties (IC50 = 66 nM) toen Flavopiridol werd getest in het panel van 60 menselijke tumorcellijnen van het National Cancer Institute Development Therapeutics Program. Flavopiridol induceert G1-arrest met remming van CDK2 en CDK4 in menselijke borstcarcinoomcellen op een tijd- en concentratieafhankelijke manier. Een korte behandeling met Flavopiridol (~12 uur) induceert apoptose in hematopoëtische cellijnen, waaronder SUDHL4, SUDHL6 (B-cellijnen), Jurkat en MOLT4 (T-cellijnen), en HL60 (myeloïde). In de clonogene assay fungeert Flavopiridol als een zeer krachtige cytotoxische verbinding met een gemiddelde IC70 van 8 ng/mL in 23 menselijke tumormodellen. Een recente studie toont aan dat behandeling met Flavopiridol een substantiële AKT-Ser473-fosforylering induceert in de menselijke glioblastoom T98G-cellijn.
Kinase Assay
Recombinante CDKs Kinase-reacties
De activiteiten van CDKs worden als volgt bepaald in microtiterplaten. Veertig μg Gst-Rb worden gemengd met verschillende hoeveelheden Flavopiridol (Alvocidib) HCl en ongelabeld ATP. Reacties worden vervolgens gestart door toevoeging van een ammoniumsulfaatfractie van de S100-fractie verkregen uit insectencellen die recombinante humane CDKs tot expressie brengen. De uiteindelijke reactiecondities zijn 10 mM MgCl2, 50 mM Tris-HCl (pH 7,5) en 1 mM DTT. De eindconcentratie van ATP wordt dienovereenkomstig aangepast. Radioactief gelabeld ATP wordt gebruikt als fosforyldonor. De reactie wordt na toevoeging van het enzym gedurende 2,5 minuten bij 30 °C uitgevoerd en vervolgens beëindigd met de toevoeging van EDTA. Het Gst-Rb wordt vervolgens opgevangen met glutathion-Sepharose en de ingebouwde radioactiviteit wordt bepaald door middel van vloeistofscintillatietelling.
In vivo
Bij de maximaal getolereerde dosis van 10 mg/kg/dag, oraal (p.o.) toegediend op dagen 1-4 en 7-11, veroorzaakt Flavopiridol (Alvocidib) HCl tumorregressie bij PRXF1337 en tumorstase die 4 weken aanhoudt bij PRXF1369. Na behandeling met een bolus van 7,5 mg/kg Flavopiridol (Alvocidib) HCl intraveneus (IV) of intraperitoneaal op elk van 5 opeenvolgende dagen, ondergaan 11 van de 12 subcutane (s.c.) humane HL-60-xenografts in een vergevorderd stadium volledige regressie, en blijven de dieren meerdere maanden na één kuur met Flavopiridol (Alvocidib) HCl ziektevrij. SUDHL-4 s.c.-lymfomen behandeld met een bolus van 7,5 mg/kg Flavopiridol (Alvocidib) HCl IV gedurende 5 dagen ondergaan ofwel een grote (twee van de acht muizen) of volledige (vier van de acht muizen) regressie, waarbij twee dieren meer dan 60 dagen ziektevrij blijven. De totale groeivertraging is 73,2%. Dagelijkse IV- of IP-toediening van Flavopiridol (Alvocidib) HCl resulteert in piekplasmaspiegels van ongeveer 7 µM, gevolgd door een geleidelijke daling naar ongeveer 100 nM in 8 uur.
