Beschrijving (Z)-4-Hydroxytamoxifen ((Z)-4-OHT, trans-4-Hydroxytamoxifen, 4-Monohydroxytamoxifen) is een selectieve modulator van estrogen receptor (SERM) met een IC50 van 3,3 nM voor de [3H]oestradiolbinding aan de oestrogeenreceptor.

In vitro

Deze verbinding is een partiële oestrogeen die bindt aan oestrogeenreceptoren met een relatieve bindingsaffiniteit (RBA) van 2,9 en de groei van de MCF-7 menselijke borsttumorcelfamilie in vitro blokkeert.
In Vivo

(Z)-4-Hydroxytamoxifen is een partiële oestrogeen met anti-oestrogene eigenschappen, en het cis-isomeer is minder potent dan het trans-isomeer. Deze verbinding remt de werking van estradiolbenzoaat en vermindert de uterine groeisnelheid in vivo.

    MCF-7 cells are incubated at 37℃ for 50 min with 1 nM [3H]estradiol in the absence or presence of increasing amounts (0.1 nM - 10 μM) of (Z)-4-Hydroxytamoxifen or unlabeled estradiol (control). Bound compounds are then extracted with ethanol, and the amounts of estrogen receptor bound [3H]estradiol are measured.
