Biologische activiteit

Beschrijving YK-4-279 is een krachtige remmer van EWS-FLI1-binding aan RNA-helicase A (RHA).
Doelen
RHA
In vitro YK-4-279 elimineert cycline D-niveaus door de interactie van EWS-FLI1 met RHA te blokkeren in EWS-FLI1-bevattende TC32-cellen. Deze verbinding remt ook specifiek de groei van ESFT-cellen en induceert apoptose. Het remt ook de biologische activiteit van ERG en ETV1 in fusiepositieve prostaatkankercellen, en vermindert verder de celmotiliteit en invasie.
In Vivo In vivo remt YK-4-279 (1,5 mg/dosis i.p.) de groei van ESFT-xenografttumoren. In een muismodel voorkomt deze verbinding selectief de groei en metastase van prostaatkanker in fusiepositieve LNCaP-luc-M6-tumoren.

Klantproductvalidatie

YK-4-279-induced cell apoptosis of NB cells by Western blot assay. SH-SY5Y cells were treated with YK-4-279 (0, 0.1 μM, 0.3 μM, 1 μM, 3 μM) for 24 h. Whole cell lysates were subjected to SDS-PAGE and immunoblotted with antibodies against PARP and Caspase 3 to detect apoptosis. β-actin was detected as loading control. YK-4-279-induced apoptosis of NB cell line SH-SY5Y by FACS. Cells were treated with YK-4-279 (0, 1 μM, 3 μM) for 24 h, and then stained by PI and analyzed by FACS.
