Biologische activiteit

Beschrijving UMI-77 is een selectieve Mcl-1 remmer met een Ki van 490 nM, die selectiviteit vertoont ten opzichte van andere leden van de Bcl-2 familie.

In vitro UMI-77 verstoort effectief de interacties tussen BL-Noxa en cellulaire Mcl-1, evenals Mcl-1/Bax eiwit-eiwitinteracties. Deze verbinding remt de groei van pancreaskankercellen met IC50-waarden van respectievelijk 3,4, 4,4, 12,5, 16,1 en 5,5 μM voor BxPC-3-, Panc-1-, MiaPaCa-2-, AsPC-1- en Capan-2-cellen. Het induceerde apoptosis in pancreaskanker door activering van de intrinsieke apoptotische route en/of Bax-conformatiële verandering.
In Vivo In een BxPC-3 xenograft muismodel vertoont UMI-77 (60 mg/kg i.v.) antitumoractiviteit als enkelvoudig middel zonder normale weefsels te beschadigen.

  • Fluorescentiepolarisatie (FP)-gebaseerde bindingsassays

    Gebaseerd op de Kd waarden zijn de concentraties van de eiwitten die gebruikt worden in de competitieve bindingsexperimenten 90 nM voor Mcl-1, 40 nM voor Bcl-w, 50 nM voor Bcl-xL, 60 nM voor Bcl-2, en 4 nM voor A1/Bfl-1. De fluorescerende probes, Flu-BID en FAM-BID, zijn vastgesteld op 2 nM voor alle assays behalve voor A1/Bfl-1, waar FAMBID op 1 nM wordt gebruikt. 5 μL van deze verbinding in DMSO en 120 μL van eiwit-/probecomplex in de assaybuffer (100 mM kaliumfosfaat, pH 7,5; 100 μg/ml runderserumalbumine; 0,02% natriumazide) worden toegevoegd aan assayplaten (Microfluor 2Black), 3 uur geïncubeerd bij kamertemperatuur en de polarisatiewaarden (mP) worden gemeten bij een excitatiegolflengte van 485 nm en een emissiegolflengte van 530 nm met behulp van de Synergy H1Hybrid plaatlezer. IC50-waarden worden bepaald door niet-lineaire regressiepassing van de competitiekurven.

    Human pancreatic cancer cell lines AsPC-1, BxPC-3, and Capan-2 are cultured in RPMI-1640 medium, whereas Panc-1 and MiaPaCa are cultured in Dulbeccos' Modified Eagle's Medium (DMEM), all supplemented with 10% FBS. The cell growth inhibition after treatment with increasing concentrations of this compound is determined by WST-8 assay.
