Beschrijving Tirzepatide (LY3298176, GIP/GLP-1 RA, TZP) natrium is een dubbele GIP/GLP-1-receptoragonist. Tirzepatide induceert differentieel de internalisatie van de GIP- en GLP-1-receptoren met EC50-waarden van respectievelijk 18,2 nM en 18,1 nM.

In vitro

In HEK293-cellen induceert Tirzepatide de internalisatie van de GIPR en heeft het een neiging tot partieel agonisme, en dit weerspiegelt uiteindelijk een signaalvoorkeur voor de cAMP-route versus β-arrestine-recrutering.
In Vivo

LY3298176 vertoont glucoseafhankelijke insulinesecretie en verbetert de glucosetolerantie door in te werken op zowel GIP- als GLP-1-receptoren bij muizen. Chronische behandeling met LY3298176 resulteert in een significante dosisafhankelijke afname van het lichaamsgewicht die meer uitgesproken is dan die waargenomen bij semaglutide.
