 2.500mg/ml (10.07mM) Taking the 1 mL working solution as an example, add 50 μL of 50 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to clarify; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. Beschrijving Thiamet G is een potente, selectieve O-GlcNAcase-remmer met een Ki van 21 nM, terwijl het een 37.000-voudige selectiviteit vertoont ten opzichte van humaan lysosomaal –hexosaminidase.

In vitro In NGF-gedifferentieerde PC-12-cellen verhoogt remming van O-GlcNAcase door Thiamet G de cellulaire niveaus van O-GlcNAc met een EC50 van ongeveer 30 nM. Deze verbinding (100 mM) vermindert tau-fosforylatie met respectievelijk ongeveer 2,1-voudig, 2,7-voudig, 1,2-voudig en 1,3-voudig voor Ser396, Thr231, Ser422 en Ser262. Het (12,5 nM en 25 nM) verbetert de p38-fosforylatie significant door O-GlcNAcylatie in mesangiale cellen te verhogen. In O-GlcNAc transferase- of O-GlcNAcase-gain-of-function-cellen herstelt deze verbinding de assemblage van de spoel en redt het gedeeltelijk de histonfosforylatie.
In Vivo Bij ratten overschrijdt Thiamet G (50 mg/kg), intraveneus toegediend, de bloed-hersenbarrière en resulteert vervolgens in een toename van de O-GlcNAc-niveaus in de hersenen op een dosis- en tijdafhankelijke manier, en een vermindering van tau-fosforylatie in de rattenhersenen. Deze verbinding is ook oraal biologisch beschikbaar. O-GlcNAc-accumulatie geïnduceerd door deze chemische stof stimuleert chondrogene differentiatie bij C57/bl-muizen door de genexpressie van differentiatie-markers te verhogen, evenals de activiteit van MMP-2 en -9.
