 4.7mg/ml (2.76mM) Taking the 1 mL working solution as an example, add 50 μL of 94 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to clarify; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. The mixed solution should be used immediately for optimal results.

Biologische activiteit

Beschrijving Tannic acid (gallotanninezuur), een polyfenolische verbinding, is een CXCL12/CXCR4-remmer met anti-angiogene, ontstekingsremmende en antitumoractiviteit.
Doelen
CXCR4
In vitro Tannic acid remt bij niet-toxische concentraties specifiek de CXCL12-geïnduceerde migratie van menselijke monocyten (IC50, 7,5 R g/ml), maar remt geen CCL2-, CCL3-, CCL5-, formylmethionylleucylphenylalanine (fMLP)- of C5a-geïnduceerde migratie. Deze verbinding blokkeert opmerkelijk de binding van CXCL12 aan THP-1-cellen (IC50, 0,36 R g/ml). Het remt ook de CXCL12-geïnduceerde, maar niet de epidermale groeifactor-geïnduceerde, migratie van MDA 231 borsttumorcellen. Bovendien remt 0,5 R g/ml van deze chemische stof selectief de CXCL12-gemedieerde, maar niet de basic fibroblast growth factor- of endotheelcelgroeisupplement-gemedieerde, capillaire buisvorming van runderaorta-endotheelcellen.
In Vivo Toegediend in drinkwater verhoogt deze verbinding de overlevingskans van muizen met syngene tumoren.

    Freshly isolated human monocytes, THP-1 cells or BAEC cells, are plated into 96-microwell plates at 1 × 105 cells/well in the presence of tannic acid at a final concentration of 100 μg/ml. Cell viability is determined by [3H]thymidine incorporation (THP-1 cells and BAEC cells) or trypan blue exclusion (human monocytes), 24 h after coincubation at 37°C in humidified air with 5% CO2.
