Biologische activiteit

Beschrijving T-5224 is een selectieve remmer van transcriptiefactor c-Fos/activator protein (AP)-1 met ontstekingsremmende effecten. T-5224 remt specifiek de DNA-bindingsactiviteit van c-Fos/c-Jun en de IL-1β-geïnduceerde opregulatie van Mmp-3, Mmp-13 en Adamts-5 transcriptie.

In vitro

T-5224 remt matrix-afbrekende MMPs en inflammatoire cytokines waaronder interleukine 1β. Deze verbinding vermindert de hoeveelheden inflammatoire cytokines en MMPs in vitro in synoviaal cel- en chondrocytenculturen.
In Vivo

T-5224 is een remmer van c-Fos/activator protein-1 (AP-1) die type II collageen-geïnduceerde artritis voorkomt door de hoeveelheden inflammatoire cytokines, MMPs en anti-tumor necrosefactor α in vivo in sera en gewrichten te verminderen.

    Reporter assay. NIH/3T3 cells are transiently transfected with the luciferase reporter plasmids pAP-1-Luc(×7 TGACTAA), pNF-kB-Luc(×5 TGGGGACTTTCCGC) or control phRL-TK, and cultured overnight. Cells are incubated in 0.5% FBS/DMEM containing T-5224 for 1 h, and then stimulated with PMA (10 ng/ml) or TNFa (10 ng/ml), and then cultured for 3 h, followed by measurement of lysate by using dual-luciferase reporter assay system.
