 5mg/ml (9.58mM) Taking the 1 mL working solution as an example, add 50 μL of 100 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to clarify; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. Biologische activiteit

Beschrijving Sodium Tauroursodeoxycholate (TUDC) is een wateroplosbaar galzout, gebruikt voor de behandeling van galstenen en levercirrose.
In vitro In vitro vertoont Tauroursodeoxycholate beschermende en omkerende effecten op 17βEG-geïnduceerde stoornissen van de cVA-of-CLF. In Mz-ChA-1 cholangiocarcinoomcellen remt Tauroursodeoxycholate de celgroei via Ca2+-, PKC- en MAPK-afhankelijke routes. In geïsoleerde rattenhepatocyten remt Tauroursodeoxycholate galzuur-geïnduceerde apoptose via een β1-integrine-gemedieerde vorming van cAMP.
In Vivo Bij phalloidine-geïnduceerde cholestase-ratten onderdrukt Tauroursodeoxycholate (360 μmol/kg, i.v.) significant de afname van de galstroom en de toename van de serum alkaline fosfatase, leucine aminopeptidase en glutaminezuur pyruvaat transaminase activiteiten, cholesterol-, fosfolipide- en galzuurconcentraties.

    After trypsinization, Mz-ChA-1 cells are suspended in CCM at 5 × 104 cells/mL, and 200 μL of the cell suspension are seeded into flat-bottomed 96-well plates. After an initial incubation step of 4 h at 37°C in a 5% CO2 incubator, the cells are incubated at 37°C with TUDCA (0.2–200 μM). After incubation for 24–96 h, [3H]thymidine is put into each well at 5.0 μCi/mL and incubated for additional 4 h. [3H]thymidine incorporation is measured by a scintillation counter. At the end of each incubation period, cell viability is evaluated by trypan blue exclusion.
