Biologische activiteit

Beschrijving Sodium oxamate (SO, Aminooxoacetic acid, Oxamic acid) is een remmer van lactate dehydrogenase (LDH) die specifiek LDH‑A remt. Deze verbinding induceert G2/M-celcyclusarrest via downregulatie van de CDK1/cyclin B1-route en bevordert apoptosis door verbetering van mitochondriale ROS-generatie.

Sodium oxamate, een specifieke remmer van LDH-A, verhoogt de onderdrukkende effecten van PARP-remmers op eierstokkanker zonder BRCA-mutaties, wat de remmende effecten van PARP-remmers op wild-type BRCA eierstokkankercellen opmerkelijk bevordert.
In Vivo

Sodium oxamate vermindert de groei van darmkanker bij CRC-muizen, door de neerwaarts gereguleerde MMR-functionele eiwitten te herstellen.

  • Methode

    Cell proliferation is evaluated using a Cell Counting Kit-8 (CCK-8). SKOV3 cells and A2780 cells are seeded in a 96-well culture plates at a density of 2×105 cells/well and incubated with different concentrations of the drugs (this compound, AG14361) for different times. Detection reagent is added to the cells, and the luminescence signals are determined.
