SAG (Smoothened Agonist) is een potente agonist van Smoothened (Smo). Smoothened Agonist induceert firefly luciferase expressie in Shh-LIGHT2 cellen met een EC50 van 3 nM.

Smoothened Agonist (SAG) kan het antagonisme van glucocorticoïden (GC's) op de Shh-gestuurde proliferatie van granulaire neuronvoorlopers (CGNP) tegengaan.

In Vivo

Systemische toediening van Smoothened Agonist (SAG) voorkomt de neurotoxische effecten van glucocorticoïden. Deze verbinding kan de bloed-hersenbarrière passeren om Shh transcriptionele targets in vivo te activeren. Bij de behandelingsdosis voorkomt het effectief GC-geïnduceerde neonatale cerebellaire ontwikkelingsstoornissen.
