Sitravatinib (MGCD516, MG-516) is een nieuwe kleine molecuulremmer die zich richt op meerdere RTK's die betrokken zijn bij het aandrijven van de groei van sarcoomcellen, waaronder c-Kit, PDGFRβ, PDGFRα, c-Met en Axl.

In vitro

Sitravatinib (MGCD516) is een orale, potente kleine molecuulremmer van een nauw verwant spectrum van RTK's, waaronder RET, de split RTK's (VEGFR, PDGFR en KIT), TRK family, DDR2, MET en AXL. Behandeling met deze verbinding resulteerde in een significante blokkade van fosforylering van potentiële driver RTK's en induceerde potente anti-proliferatieve effecten in vitro.

In Vivo

Sitravatinib (MGCD516) heeft antitumoractiviteit aangetoond in niet-klinische kankermodellen die genetische veranderingen van zijn doelwitten vertonen, waaronder herrangschikking van RET, NTRK of CHR4q12-amplificatie. Behandeling van tumorxenografts in vivo met deze verbinding resulteerde in een significante onderdrukking van tumorgroei.
