Biologische activiteit

Beschrijving RSL3 ((1S,3R)-RSL3) is een ferroptosis-activator op een VDAC-onafhankelijke manier, met selectiviteit voor tumorcellen die oncogeen RAS dragen. RSL3 bindt, inactiveert GPX4 en medieert zo GPX4-gereguleerde ferroptosis.

In vitro Ferroptosis-inducerende verbindingen inactiveren GPX4 door directe binding of door glutathion uit te putten. Na binding aan GPX4 inactiveert deze verbinding GPX4 om ROS-productie uit lipidperoxidatie te induceren. Het vermogen om synthetische letaliteit met oncogeen RAS te induceren is snel en vrij potent. Deze verbinding remt de groei van BJ-TERT/LT/ST/RASV12 en DRD-cellen tot 10 ng/ml en begon gevoelige cellen te doden al 8 uur na behandeling.
In Vivo Prostaglandine-endoperoxide synthase (PTGS) is het sleutelenzym in de prostaglandinebiosynthese. Er zijn twee isozymen van PTGS: een constitutieve PTGS1 en een induceerbare PTGS2. PTGS2, dat cyclooxygenase-2 (COX-2) codeert, wordt significant opgereguleerd na behandeling met RSL3 en deze verbinding bij muizen.

    TERT/LT/ST/RASV12 cells are seeded in 10 cm dishes and treated with 1 µM staurosporine, 10 µg/ml erastin, 20 µg/ml RSL5, and 1 µg/ml RSL3 for 16 hr. Both dying cells and live cells in each 10 cm dish are harvested and collected in the same 15 ml tubes by centrifuging cell suspension at 1000 rpm for 5 min.

ATG3 is required for ferroptosis. ATG3KO MEFs, or ATG3KO cells reconstituted with ectopic ATG3 expression, were treated as indicated for 12 h. Cell death was determined by PI staining coupled with flow cytometry. ATG3 expression was confirmed by immunoblotting. erastin: 1 μM; RSL3: 1 μM.
