 5.000mg/ml (4.49mM) Taking the 1 mL working solution as an example, add 50 μL of 100 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to clarify; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL.     Antiviral activity assays for HIV-1 and HBV are performed at Southern Research Institute. Pibrentasvir (ABT-530) is tested in an HIV-1 antiviral cytoprotection assay using CEM-SS cells and the IIIB strain of HIV-1. Briefly, virus and cells are mixed in the presence of this compound or zidovudine (AZT; positive control) and incubated for 6 days. The titers of the virus are determined beforehand such that the virus-infected control wells exhibited approximately 85% to 95% loss of cell viability due to virus replication. Therefore, antiviral effect or cytoprotection is observed when it prevented virus replication. Six days after infection, 20 to 25 μl of MTS reagent is added per well, and the microtiter plates are then incubated for 4 to 6 h to assess cell viability. Plates are read spectrophotometrically at 490/650 nm with a Molecular Devices Vmax or SpectraMax Plus plate reader.
