Biologische activiteit

Beschrijving PFK15 (PFK-015) is een potente en selectieve 6-fosfofructo-2-kinase (PFKFB3) remmer met een IC50 van 207 nM.

In vitro PFK15 produceert potente groeiremming in een reeks kankercellen. In Jurkat T-cel leukemiecellen en H522 longadenocarcinoomcellen vermindert deze verbinding ook F26BP, glucoseopname en intracellulair ATP-niveau.
In Vivo In vivo heeft PFK15 adequate farmacokinetische eigenschappen. Deze verbinding (25 mg/kg i.p.) onderdrukt de groei, metastatische verspreiding en glucosemetabolisme van LLC-tumoren in syngene muizen. In drie humane xenograftmodellen van kanker in athymische muizen levert het ook antitumorale effecten op die vergelijkbaar zijn met goedgekeurde chemotherapeutische middelen.

  • Recombinant PFKFB3-assay

    Kinase reacties worden uitgevoerd door 13 ng recombinant humaan PFKFB3-eiwit te incuberen in een reactiemix die 10 μmol/L ATP, 10 μmol/L F6P, en ofwel dimethylsulfoxide (DMSO) voertuigcontrole, 3PO, of PFK15 bevat gedurende 1 uur bij kamertemperatuur. Kinase activiteit wordt gemeten met de Adapta Universal Kinase Assay volgens de fabrikant

    Viability is determined using trypan blue exclusion. Cells were incubated in 20% trypan blue for 5 minutes. Cells excluding trypan blue are counted using a standard hemocytometer to determine the total number of viable cells. Experiments are conducted in triplicate.
