Beschrijving Osimertinib mesylate (AZD9291) is de mesylaatvorm van osimertinib, een oraal, derdegeneratie epidermal growth factor receptor (EGFR) tyrosinekinaseremmer (TKI) medicijn.

Osimertinib-resistente longadenocarcinoom (LUAD) cellen expresseren een hoger niveau van METTL7B in vergelijking met Osimertinib-gevoelige cellen. Knock-down van METTL7B door GNC-siRNA maakte LUAD-cellen opnieuw gevoelig voor osimertinib.
In Vivo

METTL7B is significant verhoogd in zowel mRNA- als proteïneniveau in osimertinib-resistente xenografts, vergeleken met die in osimertinib-gevoelige xenografts. In het LUAD CDX-muismodel onderdrukt combinatietherapie met GNC-siMETTL7B (6 mg siRNA per muis equivalent) de tumorgroei significant in vergelijking met behandeling met osimertinib (30 mg/kg) alleen.

    Cells are plated at a density of 3,000 cells/well in 96 well plates. Osimertinib is added to the medium and treated for 72 h. Cell viability assays are performed by Cell Counting Kit-8.
