Beschrijving

Nigericin sodium salt is een antibioticum afgeleid van Streptomyces hygroscopicus dat werkt als een H⁺, K⁺ en Pb²⁺ ionofoor. Nigericin kan het NLRP3 inflammasoom activeren om pro-inflammatoire en immunostimulerende processen te induceren.

In vitro model van tumorheterogeniteit de antitumorale effecten van Nigericin (NIG) dat werkt als een H⁺, K⁺ en Pb²⁺ ionofoor. NIG verminderde de levensvatbaarheid en clonogeniteit van H460-cellen, evenals hun vermogen om te migreren en te invaderen .

Met behulp van een combinatie van live-cell imaging en biochemische benaderingen tonen we aan dat prototypische NLRP3-stimuli, het kaliumionofoor nigericin, of millimolaire concentraties ATP zowel NLRP3-onafhankelijke als -afhankelijke effecten uitoefenen op macrofagen die resulteren in cytokineremissie en celdood.
