 2.800mg/ml (10.08mM) Taking the 1 mL working solution as an example, add 50 μL of clarified DMSO stock solution of 56 mg/ml to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to clarify it; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. The mixed solution should be used immediately for optimal results.

Beschrijving Necrostatin 2 racemate (Nec-1s, 7-Cl-O-Nec1, Necrostatin-1 stable, Necrostatin 1S) is een stabiele variant van Necrostatin-1. Deze verbinding is een specifiekere RIPK1-remmer zonder het IDO-targeting effect, met een >1000-voudige selectiviteit voor RIPK1 dan voor enige andere kinase van de 485 menselijke kinasen.

Necrostatin 2 racemate (Nec-1s) is een krachtige remmer van RIPK1 en cellulaire necroptose, terwijl het geen IDO-remmende activiteit heeft.
In Vivo

Necrostatin 2 racemate (Nec-1s) is effectief in het verminderen van hersenletsel. Het is een superieure remmer die geschikt is voor gebruik in vivo en geen paradoxaal sensibiliserend effect heeft bij TNF-geïnduceerde dodelijkheid.

Deze verbinding bezit verschillende voordelige farmacokinetische en farmacodynamische eigenschappen in vergelijking met Nec-1. Het veiligheidsprofiel is ook superieur aan dat van Nec-1 voor verminderde in vivo en in vitro toxiciteit.
