ML348 (GNF-Pf-1127) is een krachtige en selectieve APT1 (Acylproteïne thioesterase 1)/lysophospholipase 1 (LYPLA1) remmer met Ki-waarden van respectievelijk 280 nM en >10 μM voor APT1 en APT2.

ML348 verhoogt de hersenpalmitoylering en remt het palmitinezuur-verwijderende enzym acyl-proteïne thioesterase 1 (APT1), herstelt axonaal transport, synapshomeostase en overlevingssignalering tot wildtype-niveaus zonder toxiciteit, en verhoogt ook BDNF-trafficking in humane HD-geïnduceerde pluripotente stamcel-afgeleide corticale neuronen.

In Vivo

Bij HD knock-in muizen passeert ML348 efficiënt de bloed-hersenbarrière om palmitoyleringsniveaus te herstellen en neuropathologie, motorische tekorten en angstig-depressief gedrag om te keren.

For the fractionation experiments, we plated cortical neurons on 10-cm dishes and we treated cells for 4 hours with 1 μM this compound before lysis. For neurons within microchambers, 1 μM this chemical is administered within each compartment of the microfluidic device beginning on days in vitro (DIV)2 and continuing daily until DIV12, while for BDNF pHluorin analysis, compartments are treated for 4 hours with 1 μM this compound before acquisition.
