 0.150mg/ml (0.42mM) Taking the 1 mL working solution as an example, add 50 μL of 3 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to clarify; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. Beschrijving MKC8866 (IRE1-IN-8866), een salicylaldehyde-analoog, is een specifieke IRE1α RNase-remmer met een IC50 van 0,29 μM voor menselijke IRE1α in vitro.

MKC8866 is geoptimaliseerd en verfijnd uit een familie van IRE1α-specifieke endoribonuclease-remmers verkregen uit een chemische bibliotheekscreening. Deze verbinding onderdrukt effectief de door IRE1α gemedieerde XBP1-splicing in PCa-cellen en heeft gunstige farmacokinetische en farmacodynamische eigenschappen.
In Vivo

MKC8866 is een IRE1α RNase-specifiek remmermolecuul dat significante therapeutische activiteit vertoont in verschillende preklinische modellen van PCa in vivo. Deze verbinding synergeert met klinische PCa-geneesmiddelen in vivo.
