Beschrijving ME0328 is een krachtige en selectieve PARP-remmer met een IC50 van 0,89 μM voor PARP3, ongeveer 7 keer selectiever dan PARP1.

In vitro ME0328 is oplosbaar, celdoorlaatbaar en metabolisch stabiel in humane levermicrosomen en rattenhepatocyten. Deze verbinding (10 μM) resulteert in een significante vertraging van de γH2AX-foci-resolutie door ARTD3 in A549- en MRC5-cellen te beïnvloeden zonder significante toxiciteit.

  • Enzymatische Assays

    Eiwit-ADP-ribosylering wordt gemeten met behulp van hexahistidine-getagde ARTD-eiwitten en recombinante histoneiwitten die zijn vastgelegd op 96-wells Ni2+-chelerende platen (5-PRIME). ADP-ribosyleringsreacties worden geïnitieerd door toevoeging van NAD+ (2% gebiotinyleerd), en gemodificeerde reactieproducten worden gedetecteerd door chemiluminescentie. Km-waarden worden geschat met behulp van grafieken van initiële snelheden versus NAD+-concentraties en lineaire curvefitting met GraphPad Prism. Alle verbindingen worden opgelost in dimethylsulfoxide (DMSO) tot een stockconcentratie van 50 mM. Experimenten om IC50-waarden te bepalen, worden uitgevoerd met deze verbindingen in concentraties tussen 10 nM en 450 μM met een DMSO-concentratie van 1% (v/v). Metingen worden uitgevoerd bij een NAD+-concentratie onder Km voor elke transferase. IC50-waarden worden geschat met behulp van curvefitting met GraphPad Prism. De gerapporteerde waarden vertegenwoordigen gemiddelden ± SE van de fits van de curven, gebaseerd op dubbele of drievoudige experimenten, elk bepaald op basis van drie replicaten.

    Compound cytotoxicity in A549 and MRC5 cells is evaluated using WST-1 assays. A549 cells are cultured in Dulbecco’s Modified Eagle’s Medium supplemented with 10% fetal calf serum (FCS), penicillin, and streptomycin. MRC5 cells are cultured in Minimal Essential Medium supplemented with 10% FCS, penicillin, streptomycin, and l-glutamine. Both cell lines are maintained in a humidified incubator at 37°C and 5% CO2.

Combination drug treatment increased multinucleated giant cells via spindle microtubule alteration and cell cycle arrest. Immunofluorescent staining of BT-20 cell line after treatment with vinorelbine (NVB) or vinorelbine in combination with 5 μM of either ME0328 (NVB + ME) or olaparib (NVB + Olaparib) for 24 h. Cells were stained with Dapi for DNA (blue). α-tubulin for microtubules (green) and the merge with a 100× objective. Abundant multinucleated giant cells observed in cells treated with NVB + ME and NVB + Olaparib comparing with NVB alone.
