Beschrijving LY2409881 is een krachtige en selectieve IKK2-remmer met een IC50 van 30 nM, >10-voudige selectiviteit ten opzichte van IKK1 en andere veelvoorkomende kinasen.

In vitro LY2409881 remt constitutief geactiveerd NF-ƘB en veroorzaakt concentratie- en tijd-afhankelijke groeiremming en apoptose in diffuse grote B-cel lymfoom (DLBCL) cellen. In de eierstokkankercellijn SKOV3 vertoont deze verbinding matige cytotoxiciteit. Het is synergetisch met doxorubicine en cyclofosfamide in SUDHL2-cellen bij het remmen van de celgroei, maar niet in LY1-cellen. In zowel SUDHL2- als LY1-cellen is deze chemische stof synergetisch met de histone deacetylase (HDAC)-remmer romidepsin bij het remmen van de celgroei.
In Vivo In het SCID-beige xenograftmuismodel remt LY2409881 (50, 100 en 200 mg/kg, i.p.) de tumorgroei significant.

    Cytotoxicity is evaluated using the CellTiter-Glo Reagent according to the manufacturer's manual. Experiments are carried out in 96-well plates, with each treatment in triplicate. Samples are taken at typically 24, 48, and 72 hours after treatment. Cytotoxicity is expressed by the decreasing percentage of live cells in each treatment relative to the untreated control from the same experiment. IC50 for each cell line is calculated using the CalcuSyn Version 2.0 software.
