Biologische activiteit

Beschrijving LY2090314 is een potente GSK-3-remmer voor GSK-3 R R/ R R met een IC50 van 1,5 nM/0,9 nM; kan de werkzaamheid van op platina gebaseerde chemotherapieregimes verbeteren. Deze verbinding is zeer selectief ten opzichte van GSK3, zoals aangetoond door zijn selectiviteit in verhouding tot een groot panel van kinasen.

In vitro

LY2090314 remt selectief de activiteit van GSK-3 door de ATP-binding te onderbreken. Deze verbinding is in staat -catenine te stabiliseren. Het vertoont beperkte werkzaamheid als monotherapie. Deze chemische stof verhoogt de werkzaamheid in solide tumor kankercellijnen in vitro.

LY2090314 decreased GSK-3 phosphorylation and attenuate apoptosis inhibitor expression. a Western blots of all 3 cell lines (NGP, SK-N-AS, SH-SY-5Y), showing decreased phosphorylation of GSK-3α at Tyr279 compared to GSK-3β phosphorylation at Tyr216 as well as decrease in inactive phosphorylation ser 9 and 21. However, there is minimal to no decrease in phosphorylation of active Akt at ser 473. b Western blot analysis showed there is an increase in pro-apoptotic markers such as cleaved PARP and cleaved caspase-3, and a decrease in anti-apoptotic markers, cyclin D1, Mcl-1, and survivin.
