Beschrijving Inavolisib (GDC-0077, RG6114, RO-7113755) is een krachtige selectieve remmer van PI3K alpha (PI3K ) met een IC50 van 0.038 nM. GDC-0077 is >300-voudig selectiever voor PI3K alpha dan voor de andere klasse I PI3K-isoformen (beta, delta en gamma) en >2000-voudig selectiever dan voor PIK-familieleden. GDC-0077 bindt aan de ATP-bindingsplaats van PI3K en remt de fosforylering van PIP2 tot PIP3.

In Vivo

OG-L002 (6 tot 40 mg/kg) onderdrukt primaire HSV-infectie op een dosisafhankelijke manier in een muismodel. Bovendien onderdrukt deze verbinding ook de reactivering van HSV uit latentie in een muizenganglion-explantatiemodel.

    Inavolisib (GDC-0077) was evaluated using an ATP-based cell viability assay in colon cancer cell lines. Western blot analysis measured p110α protein levels and pAKT signaling in these cell lines treated with this compound at gradient concentrations for 24 hours.
