Biologische activiteit

Beschrijving H-151 is een zeer krachtige en covalente remmer van STING met opmerkelijke remmende activiteit, zowel in menselijke cellen als in vivo.

In Vivo

Na intraperitoneale toediening bereikt H-151 effectieve systemische niveaus, vertoont het een korte halfwaardetijd in het serum en vormt het een adduct met mmSTING. Met name de voorbehandeling met deze verbinding vermindert de systemische cytokineresponsen bij met CMA behandelde muizen aanzienlijk. Bovendien vertoont het een opmerkelijke werkzaamheid bij Trex1−/−-muizen die een bioluminescente IFNβ-reporter tot expressie brachten, wanneer het gedurende één week werd toegediend.
