Biologische activiteit

Beschrijving Elamipretide (MTP-131, SS-31) is een cytochrome c peroxidase-remmer. Elamipretide verbetert mitochondriale disfunctie, synaptische en geheugenstoornissen veroorzaakt door lipopolysaccharide bij muizen.

In vitro

Elamipretide (SS-31, MTP-131) bindt met hoge affiniteit aan cardiolipine, een anionisch fosfolipide dat tot expressie komt op de binnenste mitochondriale membraan en dat nodig is voor de vorming van cristae. Bovendien remt het complex van deze verbinding met cardiolipine de cytochroom c peroxidase-activiteit, die cardiolipineperoxidatie katalyseert en resulteert in mitochondriale schade tijdens ischemie, door het heemijzer te beschermen.
In Vivo

Voorbehandeling van ratten met Elamipretide (SS-31, MTP-131) beschermt cristae-membranen tijdens renale ischemie en voorkomt mitochondriale zwelling. Snel herstel van ATP bij reperfusie leidt tot snel herstel van ATP-afhankelijke processen, zoals herstel van het actine-cytoskelet en celpolariteit. Snel herstel van ATP remt ook apoptose, beschermt de tubulaire barrièrefunctie en vermindert renale disfunctie.

  • Methode

    Intracellular targeting of Elamipretide (SS-31, MTP-131) is determined by incubating feline kidney cells, grown in biotin-free media for 24 hours, with biotinylated compound for 1 hour. Cells are then fixed with 4% paraformaldehyde, permeabilized with 0.1% triton, and treated with Streptavidin-AlexaFluor594 for 30 minutes at room temperature. Fluorescence staining is observed by wide-field fluorescent microscope.
