Cyclosporin A

Cyclosporin A bindt aan cyclofiline (immunofiline) in T-cellen[1], vormt een cyclofiline-dit-verbinding-complex dat vervolgens bindt aan en calcineurin remt. [2] Deze chemische stof remt calcineurin met een IC50 van 7 nM[3] en blokkeert vervolgens de nucleaire translocatie van NF-AT. [4] Het beïnvloedt ook mitochondriën door het openen van de MTP (mitochondriale permeabiliteitsovergangsporie) te voorkomen met een IC50 van 39 nM. [5]

Cyclosporin A is een immunosuppressief middel dat effectief is na parenterale of orale toediening bij muizen, ratten en cavia's. [6], wordt klinisch gebruikt bij orgaantransplantatie om afstoting te voorkomen. [7]

  Gezuiverde calcineurin en calmoduline van runderhersenen worden aangekocht. Reactiemengsels met gezuiverd enzym bevatten 100 nM calcineurin, 100 nM calmoduline en 5 μM ³²P-gelabeld fosfopeptide, in 60 μl (totaal volume) assaybuffer die 20 mM Tris (pH 8), 100 mM NaCI, 6 mM MgCl₂, 0,5 mM dithiothreitol, 0,1 mg runderserumalbumine per ml, en ofwel 0,1 mM CaCl₂ of 5 mM EGTA bevat. Reactiemengsels met cellysaten bevatten 20 μl onverdund lysaat, 5 μM ³²P-gelabeld fosfopeptide en 40 μl assaybuffer. Waar aangegeven, bevatten reactiemengsels 50 μM peptide 412 of 413 en/of 500 nM okadisch zuur, een specifieke remmer van phosphatases 1 en 2A; 500 nM okadisch zuur is voldoende voor remming van Ca²⁺-onafhankelijke phosphatases, terwijl hogere concentraties ook Ca²⁺-afhankelijke activiteit gedeeltelijk remmen. Na 15 min bij 30°C worden de reacties beëindigd door toevoeging van 0,5 ml 100 mM kaliumfosfaatbuffer (pH 7,0) met 5% trichloorazijnzuur. Vrij anorganisch fosfaat wordt geïsoleerd door Dowex kationuitwisselingschromatografie en gekwantificeerd door scintillatietelling zoals beschreven.

• Methode

  Immunosuppressive agents are dissolved in ethanol at concentrations 1000-fold more than the concentration desired for cell treatments. Cells (10⁶) are suspended in 1 ml of complete medium in microcentrifuge tubes; 1 μl of ethanol or of the ethanolic solution of Cyclosporin A is added, and the cells are incubated at 37°C for 1 hr. Cells are washed twice with 1 ml of PBS on ice and lysed in 50μl of hypotonic buffer containing 50 mM Tris (pH 7.5); 0.1 mM EGTA; 1 mM EDTA; 0.5 mM dithiothreitol; and 50 μg of phenylmethylsulfonyl fluoride, 50 μg of soybean trypsin inhibitor, 5 μg of leupeptin, and 5 μg of aprotinin per ml. Lysates are subjected to three cycles of freezing in liquid nitrogen followed by thawing at 30°C and then are centrifuged at 4°C for 10 min at 12,000×g.
