Beschrijving

CPI-455 HCl is een specifieke KDM5-remmer met een halfmaximale remmende concentratie (IC50) van 10 ± 1 nM voor full-length KDM5A in enzymatische assays, wat de globale niveaus van H3K4-trimethylering (H3K4me3) verhoogt en het aantal DTP's vermindert in verschillende kankercellijnmodellen behandeld met standaardchemotherapie of gerichte middelen.

CPI-455 heeft een verbeterde potentie tegen KDM5A, terwijl het een ~200-voudige selectiviteit voor KDM5A ten opzichte van KDM4C aantoont. CPI-455 remt KDM5A, KDM5B en KDM5C in vergelijkbare mate, maar vertoonde aanzienlijk zwakkere potentie ten opzichte van KDM4C en KDM7B (respectievelijk ~200- en 770-voudig) en geen meetbare remming van KDM2B, KDM3B of KDM6A. CPI-455-gemedieerde KDM5-remming resulteert in een dosisafhankelijke toename van globale H3K4me3 in HeLa-cellen. CPI-455 verandert specifiek de H3K4-methylering in cellen en bindt zich aan de demethylase actieve plaats.
