 2.000mg/ml (7.45mM) Taking the 1 mL working solution as an example, add 50 μL of 40 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to make it clear; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. The mixed solution should be used immediately for optimal results.

Beschrijving Ciclopirox ethanolamine (Ciclopirox olamine, HOE 296, Ciclopiroxolamine) is een breedspectrum antischimmelmiddel dat werkt als een ijzerchelaator.

Ciclopirox remt de groei van C. albicans stam SC5314 cellen aanzienlijk, met MIC80s van 1,0-2,0 μg/mL, de groei nam drastisch af bij concentraties van >0,6 μg/mL, en bijna volledige groeiremming bij een concentratie van 0,7 μg/mL, in tegenstelling tot fluconazol dat een veel breder scala aan concentraties met intermediaire remming vertoont. Net als ijzerchelaator bipyridine, vermindert Ciclopirox de celgroei door binding aan ijzerionen, wat kan worden omgekeerd door toevoeging van FeCl3. Bovendien vermindert Ciclopirox behandeling bij subinhiberende concentratie (0,6 μg/mL) slechts matig de virulentiegenen zoals genen die coderen voor uitgescheiden proteinasen of lipasen, maar leidt tot een duidelijke op- of neerwaartse regulatie van genen die coderen voor ijzerpermeases of Transmembrane Transporters (FTR1, FTR2, en FTH1), een koperpermease (CCC2), een ijzerreductase (CFL1), en een siderofoor Transporter (SIT1). Hoewel de Candida geneesmiddelresistentiegenen CDR1 en CDR2 na Ciclopirox behandeling zijn opgereguleerd, kon geen verandering in resistentie of verhoogde tolerantie worden waargenomen, zelfs niet na een incubatieperiode van 6 maanden, in tegenstelling tot fluconazol waarbij de MIC's voor cellen merkbaar toenemen na 2 maanden. Ciclopirox remt de groei van Aspergillus fumigatus stam B5233 met IC50 van 4,22 μM, krachtiger vergeleken met deferipron met IC50 van 1,29 mM.

Functies Vertoont antischimmelactiviteit via een specifiek ijzerbeperkend mechanisme, waarbij geen enkel geval van Fungal resistentie is gemeld.

    Sabouraud glucose medium (2%) is used for cell culture growth, and RPMI 2% glucose medium and 2% Sabouraud glucose medium are used for MIC determinations. For cell culture growth curves, 220 mL of 2% Sabouraud glucose medium containing different concentrations of Ciclopirox are inoculated with 105 cells/mL, and the mixture is shaken at 160 rpm and 37 °C for 1-10 hours. Growth is measured photometrically at 630 nm. FeCl3 or 2,2'-bipyridine is added to the medium at different concentrations for inhibition studie
