Beschrijving C646 is een remmer voor histonacetyltransferase en remt p300 met een Ki van 400 nM in een celvrije test. Bij voorkeur selectief voor p300 boven andere acetyltransferases. C646 induceert celcyclusarrest, apoptose en autophagy.

In vitro C646 is een remmer voor histone acetyltransferase, remt p300 met een Ki van 400 nM en is selectief ten opzichte van andere acetyltransferases. Deze verbinding produceert 86% remming van p300 in vitro bij 10 μM. Het is een klassieke reversibele p300-remmer. Deze chemische behandeling (25μM) vermindert de histon H3- en H4-acetylatieniveaus en heet TSA-geïnduceerde acetylatie in cellen op. Het (20μM) induceert apoptose in androgeengevoelige en castratie-resistente prostaatkankerzellijnen door interferentie met AR- en NF-kB-routes. Deze verbinding blokkeert dynamische acetylatie van H3K4me3 wereldwijd in muis- en vliegcellen, en lokaal over de promotor en startplaats van induceerbare genen in de muis, waardoor de associatie van RNA-polymerase II en de activering van deze genen wordt verstoord.
In Vivo C646 toegediend in de ILPFC onmiddellijk na zwakke extinctietraining verbetert de consolidatie van angstextinctiegeheugen. Deze verbinding vermindert mechanische allodynie en thermische hyperalgesie, vergezeld van een onderdrukte COX-2-expressie, in het ruggenmerg.
Functies Wordt uitgebreid gebruikt als farmacologisch probe in kankercellen. Potentieel gebruik voor prostaat- en longkanker.

Kinase-assay:[1]
  • Radioactieve test

    IC50-waarden voor de vermeende p300 HAT-remmers worden bepaald met behulp van de hierboven beschreven directe radioactieve test. Reacties worden uitgevoerd in 20 mM HEPES (pH 7.9), en bevatten 5 mM DTT, 80μM EDTA, 40μg/ml BSA, 100μM H4-15 en 5 nM p300. Vermeende remmers worden toegevoegd over een reeks concentraties, waarbij de DMSO-concentratie constant wordt gehouden (<5%). Reacties worden gedurende 10 min bij 30°C geïncubeerd en vervolgens geïnitieerd met toevoeging van een 1:1-mengsel van 12C-acetyl-CoA en 14C-acetyl-CoA tot 20 mM. Na 10 min bij 30°C worden de reacties geblust met 14% SDS (w/v). Alle concentraties worden in tweevoud gescreend. Gels worden gelopen, gewassen, gedroogd en blootgesteld aan een PhosphorImager-plaat, en de productie van Ac-H4-15 wordt gekwantificeerd om IC50's te verkrijgen.

    Histone acetylation assays in mouse cells. C3H10T1/2 mouse fibroblasts are grown in DMEM with 10% FCS at 37°C with 6% CO2. Confluent cultures are rendered quiescent in DMEM with 0.5% FCS for 18-20 hr prior to treatment. Cells are treated with the following compounds: TSA (10 ng/ml [33 nM]), C646 (25 μM), this compound (25 μM). Antibodies are used at the following concentrations: total H3 (1:10000; ab7834; Abcam); H4K12ac (1:2500; 06-761; Upstate). Rabbit anti-H3K9ac (1:10000) antibodies are generated in-house. Histones are isolated from cells by acid extraction, separated by SDS and acid-urea polyacrylamide gel electrophoresis and analyzed by western blotting.
