 1.250mg/ml (2.38mM) Taking the 1 mL working solution as an example, add 50 μL 25 mg/ml clarified DMSO stock solution to 400 μL PEG300, mix evenly to clarify it; add 50 μL Tween80 to the above system, mix evenly to clarify it; then continue to add 500 μL ddH2O to adjust the volume to 1 mL. Voorbereiden van stamoplossingen

Biologische activiteit

Beschrijving BPTES is een krachtige en selectieve Glutaminase GLS1 (KGA)-remmer met een IC50 van 0,16 μM. Het heeft geen effect op de glutamaatdehydrogenase-activiteit en veroorzaakt slechts een zeer lichte remming van de γ-glutamyltranspeptidase-activiteit.

In vitro

BPTES remt de glutaminase-activiteit die tot expressie komt in menselijke niercellen met een IC50 van 0,18 μM, en remt de glutamaatuitstroom door microglia met een IC50 van 80-120 nM. Deze verbinding vertraagt bij voorkeur de celgroei in D54-cellen met mutante IDH1. Het remt ook de glutaminase-activiteit, verlaagt de glutamaat- en α-KG-niveaus en verhoogt de glycolytische intermediairen. Deze chemische stof (10 μM) remt de celgroei van mHCC 3–4-cellen afkomstig van LAP/MYC-tumoren. Het remt ook de groei van MYC-afhankelijke P493-cellen door DNA-replicatie te blokkeren, wat leidt tot celdood en fragmentatie.

In Vivo

Bij LAP/MYC-muizen verlengt BPTES (12,5 mg/kg, i.p.) de overleving zonder significante effecten op MYC-, GLS- of GLS2-niveaus. Deze verbinding (200 μg/muis, i.p.) remt ook de tumorgroei van muizen met P493-tumorxenografts.

  • Glutaminase-remming: celvrije test

    Testplaten worden voorbereid met 2 μL testverbinding in DMSO/putje. Het enzym wordt verdund tot 1 eenheid (lever) of 0,8 eenheid (nier)/100 μL in glutaminase-analysebuffer, en 100 μL verdund enzym wordt met Multidrop aan elk putje van de analyseplaat toegevoegd. De inhoud wordt gemengd door 1 minuut op volle snelheid te schudden op TiterMix 100. De platen worden 20 minuten bij kamertemperatuur (KT) geïncubeerd om binding van testverbindingen aan glutaminase mogelijk te maken, en 50 μL glutamineoplossing (7 mM in analysebuffer) wordt met Multidrop aan elk putje toegevoegd. De inhoud wordt 30 seconden op volle snelheid geschud op TiterMix 100, en de platen worden vervolgens 60 minuten (lever) of 90 minuten (nier) bij KT geïncubeerd. Om de reacties te stoppen, wordt 20 μL HCl (0,3 N) met Multidrop aan elk putje toegevoegd en onmiddellijk gemengd door 30 seconden op TiterMix 100 te schudden. Voor kwantificering wordt glutamaat (gevormd door glutaminase-gekatalyseerde hydrolyse van glutamine) geoxideerd tot 2-oxoglutaraat door een tweede enzym, glutamaatdehydrogenase (GDH), met de gelijktijdige productie van de gereduceerde vorm van nicotinamide-adeninedinucleotide (NADH). Reductie van nitroblauw tetrazolium (NBT) in de analyseoplossing door NADH, gekatalyseerd door fenazinemethosulfaat (PMS), resulteert in de vorming van een blauwpaars formazan. De absorptie van formazan bij 540 nm is lineair evenredig met de concentratie van glutamaat tot 200 μM. NBT/GDH-reagens (50 μL) wordt met Multidrop aan elk putje toegevoegd en gemengd door 30 seconden op TiterMix 100 te schudden, en de platen worden 20 minuten bij KT geïncubeerd om kleurvorming door de GDH-reactie mogelijk te maken. De glutamaatconcentratie wordt bepaald uit de formazanconcentratie, zoals bepaald door de OD540 nm op een SpectraMax 340 af te lezen.

    Cell growth assays are carried out using alamarBlue. Cells are plated at a density of 500 cells/well in a 96-well black clear bottom plate. At 24 hrs, media is changed to the appropriate media (DMEM with 4.5 g/L, 1.5 g/L or 0.1 g/L glucose, 10% FBS, pencillin/streptomycin, and 4 mM glutamine). 48 hours after plating, compounds or DMSO are added. Media and alamarBlue is added to a volume of 200 礚 in each well. Fluorescence is measured at 48 hrs (AOA, this compound) using a Victor3 plate-reader.
