Biologische activiteit

Beschrijving Blasticidin S HCl is een nucleoside antibiotic geïsoleerd uit Stretomyces girseochromogenes. Het werkt door de hydrolyse van peptidyl-tRNA, geïnduceerd door releasefactoren, te blokkeren en remt de vorming van peptidebindingen. Het wordt gebruikt om getransfecteerde cellen te selecteren die bsr- of BSD-resistentiegenen dragen.

In vitro

Blasticidin S remt de eiwitsynthese door in te werken op de stap van peptidetransfer van peptidyl-sRNA naar inkomend aminoacyl-sRNA.

Blasticidin S remt ook de DNA/RNA Synthesis aanzienlijk, onafhankelijk van hun effect op de eiwitsynthese.

Blasticidin S doodt de cellen sneller dan G418, wat het blasticidin S-resistentiegen (bsr) tot een selecteerbare merker voor zoogdiercellen maakt.

Bovendien bindt Blasticidin S ook efficiënt Cu(II)-ionen en voorkomt het DNA van metaal-geïnduceerde schade.

In Vivo

Blasticidin S HCl is een antibiotic geïsoleerd uit Stretomyces girseochromogenes dat werkt als een eiwitsyntheseremmer. Het wordt gebruikt om getransfecteerde cellen te selecteren die bsr- of BSD-resistentiegenen dragen.
