Biologische activiteit

Beschrijving BAY 87-2243 is een krachtige en selectieve hypoxia-inducible factor-1 (HIF-1) remmer. Deze verbinding remt de activiteit van mitochondrial complex I, wat een mitofagie-afhankelijke ROS-toename teweegbrengt die leidt tot necroptose en ferroptosis. Het oefent antitumoractiviteit uit. Fase 1.

In vitro BAY 87-2243 remt HIF-1 reportergenactiviteit en CA9-eiwitexpressie met IC50 van respectievelijk 0,7 nM en 2 nM. In hypoxische longkankercellen H460 onderdrukt deze verbinding de expressie van HIF-doelgenen en remt het de accumulatie van HIF-1α-eiwit. Bij glucosegebrek remt het de celproliferatie via interferentie met de mitochondriale functie.
In Vivo In een H460-xenograftmuismodel vermindert BAY 87-2243 (4 mg/kg p.o.) het tumorgewicht, het HIF-1α-eiwit en de expressieniveaus van HIF-1-doelgenen.

    To evaluate the cytotoxicity of BAY 87-2243, 2.000 cells of the respective cell lines are seeded in 96-well plates and cultured in the appropriate growth medium containing 10% FCS. This compound at various concentrations is added at 24 h after seeding for additional 48 h and cell viability is determined using Cell Titer Glow Assay.

Fresh human PMP tissue was exposed to HIF-1α inhibitor (BAY 87-2243) under hypoxic conditions in vitro for 24 hours. MUC2 mRNA levels as measured by real-time PCR (F) and MUC2 protein expression as measured by immunofluorescence staining (G) were significantly reduced by the inhibitor; for immunofluorescence assay slides were stained with MUC2 antibody (green IF) and SYTOX Orange for nucleic acid (blue IF). Confocal images were randomly taken of 10 different fields (X 63 magnification) and analyzed using Image-pro Premier Software to quantify the average intensity of MUC2 expression.

Quiescent VSMC were pretreated with 10 or 30 μM HIF inhibitor BAY 87-2243 and then stimulated with PDGF (10 ng/mL) for 6 h. Total cell lysates were subjected to immunoblot analysis for HK2 and actin.
