Beschrijving

A-83-01 is een potente inhibitor van de TGF-eta type I receptor (ALK5-TD) met een IC50 van 12 nM. Deze verbinding inhibeert ook de transcriptie geïnduceerd door de activine/nodale type I receptor (ALK4-TD) en de nodale type I receptor (ALK7-TD) met een IC50 van respectievelijk 45 nM en 7.5 nM.Oplossingen zijn onstabiel en moeten vers bereid worden.

In vitro

A-83-01 inhibeert de transcriptionele activiteit geïnduceerd door TGF-eta type I receptor ALK-5 en die door activine type IB receptor ALK-4 en nodale type I receptor ALK-7, waarvan de kinase domeinen structureel sterk verwant zijn aan die van ALK-5. Deze verbinding is potent in de inhibitie van ALK5 en voorkomt ook fosforylering van Smad2/3 en de groeiremming geïnduceerd door TGF-eta.

In Vivo

A83-01 behandeling verhoogt significant het aantal Nkx2.5+ cardiomyoblasten bij baseline en na myocardiale schade, wat resulteert in een toename van nieuw gevormde cardiomyocyten. Deze verbinding verbetert significant de ventriculaire elastantie en slagvolume, wat leidt tot verbeterde contractiliteit na letsel.
