Beschrijving A-205804 is een potente en selectieve remmer van E-selectine- en ICAM-1-expressie met respectievelijk een IC50 van 20 nM en 25 nM.

In vitro A-205804 remt selectief de oppervlakte-expressie van de celadhesiemoleculen ICAM-1 en E-selectine in menselijke endotheelcellen. Deze verbinding remt de IL-1 l-ge duceerde expressie van E-selectine (IC50 = 20 nM) en ICAM-1 (IC50 = 10 nM) met in wezen hetzelfde potentieniveau als voor TNF l-ge duceerde expressie. Om onduidelijke redenen is het vermogen van deze chemische stof om PMA-ge duceerde E-selectine-expressie te remmen significant verminderd (IC50 > 1000 nM), maar het is een potente remmer van ICAM-1-expressie (IC50 = 40 nM) met behulp van dit prote lne-k nase C-activerende agens.

The effect of rhICAM-1 or inhibitor A-205804 on hepsin or CtsD transfected MCF-7 cells was detected. The level of E-cadherin, claudin-1, N-cadherin, vimentin and Snail1 was analyzed by western blot.
