Biologische activiteit

Beschrijving Sodium butyrate (NaB, Butanoic acid sodium salt), natriumzout van boterzuur, is een histonedeacetylase-remmer en bindt competitief aan de zinkplaatsen van klasse I en II histonedeacetylases (HDACs). Deze verbinding remt de celcyclusprogressie, bevordert differentiatie en induceert apoptosis en autophagy in verschillende soorten kankercellen.

In vitro Butyraat medieert de groeiremming van colonkankercellen en daarmee de moleculaire link tussen een vezelrijk dieet en de arrestatie van coloncarcinogenese op te helderen. Deze verbinding induceert p21 mRNA-expressie op een onmiddellijke-vroege manier, door transactivatie van een promotor cis-element(en) gelegen binnen 1,4 kb van de transcriptionele startplaats, onafhankelijk van p53-binding. Sodium butyrate, in fysiologische concentraties, induceert apoptosis in 2 adenoomcellijnen, RG/C2 en AA/Cl, en in de carcinoomcellijn PC/JW/FI. Het oefent krachtige effecten uit op een verscheidenheid aan colonmucosa-functies, zoals remming van ontsteking en carcinogenese, versterking van verschillende componenten van de colonafweerbarrière en vermindering van oxidatieve stress. Dit chemische middel remt de meeste HDAC's, behalve klasse III HDAC en klasse II HDAC6 en -10. Het is ook het meest effectieve vetzuur in het stimuleren of onderdrukken van de expressie van specifieke genen.
In Vivo Sodium butyrate verlengt de overleving significant op een dosisafhankelijke manier, verbetert het lichaamsgewicht en de motorische prestaties, en vertraagt de neuropathologische gevolgen in het R6/2 transgene muismodel van de ziekte van Huntington (HD). Deze verbinding verhoogt ook de histon- en specificiteitsproteïne-1-acetylering en beschermt tegen 3-nitropropionzuur-neurotoxiciteit.

<p>U87 cells were cultured with DMSO or 10 µM 5azadC for 72 h. For the latter, 1 µM Trichostatin A (TSA), 10 mM sodium butyrate (NaBu), 5 mM nicotinamide (NAM), or 0.5 µM apicidin were added in the last 24 h. IFNLR1 expression was determined by RT-qPCR.</p>

Effects of combination of either MS-275, apicidin, SAHA or sodium butyrate (NaB) with bortezomib on proliferation of Wp-restricted BL cells. P3HR1-c16 cells were treated with combination of NaB (0, 0.6, 1.2, 2.4, 4.8 and 9.6 mM) for 48 h. Cell proliferation was determined by MTT assay and presented as percentages of cell proliferation of treated cells compared with those of untreated cells. Synergisms of proliferation inhibition of the cells following treatment with different drug combinations were analysed by isobologram analysis.
