Beschrijving Indirubin (NSC 105327) is een krachtige cycline-afhankelijke kinasen- en GSK-3β-remmer met een IC50 van ongeveer 5 μM en 0,6 μM.

In vitro Indirubin is het werkzame bestanddeel van Danggui Longhui Wan, een mengsel van planten dat in de traditionele Chinese geneeskunde wordt gebruikt om chronische ziekten te behandelen. Indirubin remt de CDK's-activiteit met een IC50 van 2,2 - 10 μM, wat resulteert in celcyclusarrest in de G2/M-fase. Indirubin remt ook GSK-3β met een IC50 van 0,6 μM, waardoor CDK5- en GSK-3β-gemedieerde tau-fosforylering wordt afgezwakt, een proces dat overactief is bij de ziekte van Alzheimer. Het onderdrukt ook tumornecrosefactor (TNF)-geïnduceerde NF-κB-activering op een dosis- en tijdsafhankelijke manier. Indirubin onderdrukt ook de NF-κB-activering die wordt geïnduceerd door verschillende ontstekingsmiddelen en carcinogenen. Indirubin blokkeert de fosforylering en afbraak van IκBα door de activering van IκBα-kinase en fosforylering en nucleaire translocatie van p65 te remmen.
