Biologische activiteit

Beschrijving Daphnetin, een natuurlijk coumarinederivaat, is een proteïnekinaseremmer en remt EGFR, PKA en PKC met een IC50 van respectievelijk 7,67 μM, 9,33 μM en 25,01 μM. Het staat ook bekend om zijn ontstekingsremmende en antioxidante activiteiten.

In vitro Daphnetin remt niet alleen de EGF-receptor-gekatalyseerde tyrosinefosforylatie van exogeen substraat sterk, maar ook de PKA- en PKC-activiteiten. Deze verbinding remt de MCF-7 oestrogeen-responsieve humane carcinoomcellijn bij een blootstelling van 24 uur met een IC50 van 73 μM. Het verlaagt het niveau van cycline D1 zelfs bij 50 μM. Deze chemische stof beschermt de corticale neuronen tegen dexamethason-geïnduceerde vermindering van cellevensvatbaarheid op een dosisafhankelijke manier. Het remt de activiteiten van endogeen of recombinant TaPRK op een specifieke en dosisafhankelijke manier. Deze verbinding veroorzaakt een 50% remming (IC50) van de 3H-hypoxanthine-incorporatie door Plasmodium falciparum bij concentraties tussen 25 μM en 40 μM. Het remt de mitogene signalering van ERK1/ERK2.
In Vivo Daphnetin bij 140 mg/kg vermindert het baarmoedergewicht significant met 39,5%. Een toediening van 2 en 8 mg/kg van deze verbinding zou de prestaties van gestreste muizen in Morris-waterdoolhoftests en gedwongen zwemtests kunnen verbeteren. Deze chemische stof verlengt de overleving van met P. yoelli geïnfecteerde muizen significant.

  • Analyse van proteïnekinase C (PKC) en cAMP-afhankelijke proteïnekinase (PKA) activiteiten.

    PKC- en PKA-activiteiten worden gedetecteerd. Kortom, 5 ml PKC of PKA wordt gemengd met 5 ml lipidebereiding die 100 mM forbol 12-myristaat 13-acetaat, 2,8 mg/ml fosfatidylserine en Triton X-100 gemengde micellen bevat voor de PKC-analyse, of 5 ml 40 mM cAMP in 50 mM Tris-HCl, pH 7,5, voor de PKA-analyse, en 5 ml daphnetin. De reactie wordt gestart door toevoeging van 10 ml PKC-substraatoplossing die 250 mM geacetyleerd myeline basisch proteïne, 100 mM ATP, 5 mM CaCl2, 10 mM MgCl2, 20 mM Tris-HCl, pH 7,5, of 10 ml PKA-substraatoplossing die 200 mM Kemptide, 400 mM ATP, 40 mM MgCl2, 1 mg/ml BSA, 50 mM Tris-HCl, pH 7,5, en 20-25 mCi/ml [g-32P] ATP bevat. Na incubatie bij 25 °C gedurende 5 minuten wordt 20 ml van elk mengsel op een stukje fosfocellulosedisc gespot dat onmiddellijk in 1% H3PO4 wordt geplaatst. Nadat vrij [g-32P] ATP op de discs is verwijderd, wordt het peptide-ingebouwde 32P op de discs geteld in een scintillatieteller.

  • Methode

    The cytostatic effect of Daphnetin tested on the MCF-7 tumor cells is estimated using the microculture MTT assay. The assay is based on the reduction of soluble tetrazolium salt by mitochondria of viable cells. The reduced product, an insoluble purple-colored formazan, is dissolved in dimethyl sulfoxide and measured spectrophotometrically (570 nm). The amount of formazan formed is proportional to the number of viable cells. Cells (3 × 103) are seeded in each of the 96 microplate wells in a 200 μL medium containing the corresponding concentration of this compound. This compound is tested at five concentrations (12.5 μM, 25 μM, 50 μM, 100 μM, 200 μM). After 24-hour, 48-hour, and 72-hour exposure, the percentage of proliferative inhibition of treated cells is estimated against the solvent-treated control cells (PI% = [(T/C) − 1] × 100). PI = proliferation inhibition; T = treated, C = control. IC50 is calculated from the least square concentration-response regressions.
