 3.750mg/ml (11.47mM) Taking the 1 mL working solution as an example, add 50 μL of 75 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to clarify; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. Biologische activiteit

Beschrijving Clozapine (HF 1854, LX 100-129) is een atypisch antipsychoticum dat werkt als een 5-HT antagonist en wordt gebruikt bij de behandeling van schizofrenie.
Doelen

In vitro Clozapine (10, 20 mg/kg) verhoogt significant het aantal Fos-positieve neuronen in de nucleus accumbens, de mediale prefrontale cortex en de laterale septale kern. Deze verbinding induceert zif268 maar geen c-fos mRNA in het striatum van ratten, terwijl Haloperidol c-Fos-achtige immunoreactiviteit in het caudate-putamen induceert. Het is selectiever voor D4-receptoren dan voor D2-receptoren. Deze chemische stof is een gemengde maar zwakke D1/D2-antagonist. Het produceert een duidelijke facilitatie (300-400%) van NMDA-uitgelokte reacties op een concentratie-afhankelijke manier met een EC50 van 14 nM. Deze verbinding, maar geen haloperidol, produceert bursts van exciterende postsynaptische potentialen (EPSP's), die worden geblokkeerd door glutamaatreceptorantagonisten, wat suggereert dat deze EPSP's het gevolg zijn van een toenemende afgifte van exciterende aminozuren. Het is een volledige agonist bij de muscarinerge M4-receptor (EC50 = 11 nM), wat leidt tot remming van forskoline-gestimuleerde cAMP-accumulatie. Deze chemische stof antagoniseert krachtig agonist-geïnduceerde reacties bij de andere vier muscarinerge receptoren.
In Vivo Clozapine vertoont een "omgekeerde U"-vormige dosis-respons curve, waarbij het apomorfine-geïnduceerde verlies van prepulse-inhibitie (PPI) bij lage doses wordt omgekeerd, maar niet bij hoge doses bij ratten. Deze verbinding vermindert PPI onafhankelijk van apomorfinebehandeling bij ratten.
