Beschrijving Clopidogrel (SR-25990C) Bisulfate is een oraal antiplaatjesmiddel van de thienopyridineklasse.
Doelen

In vitro Clopidogrel wordt door cytochroom P450 (CYP) enzymen omgezet in zijn actieve metaboliet. Clopidogrel (1 μM) remt ook EGF-gestimuleerde EGF-receptor, PERK-expressie en celproliferatie in RGM-1-cellen (P<0.05), en veroorzaakt veel minder remming van EGF-gestimuleerde celproliferatie in EGF-receptor over-expressie RGM-1-cellen dan in RGM-1-cellen (22% vs. 32% reductie). Clopidogrel verhoogt het aantal bloedvaten, vermindert het polymorfonucleaire aantal en vermindert hechting en botverlies, en vermindert ook het aantal osteoclasten bij ratten die al dan niet parodontale reparatie ondergaan. Clopidogrel vermindert het CXCL4-, CXCL12- en PDGF-gehalte vergeleken met met zoutoplossing behandelde ratten, zonder CXCL5 te beïnvloeden.
In Vivo Clopidogrel (2 mg en 10 mg/kg/dag) vermindert significant de door zweren geïnduceerde proliferatie van maagepitheelcellen en de door zweren gestimuleerde expressies van EGF-receptor en gefosforyleerde extracellulaire signaal-gereguleerde kinase (PERK) aan de wondrand van ratten. Clopidogrel verbetert de endotheelfunctie en NO-biologische beschikbaarheid bij ratten met congestief hartfalen. Clopidogrel-behandelde ratten met congestief hartfalen (CHF) vertonen een verbeterde fosforylering van AKT en eNOS. De combinatie clopidogrel/aspirine vertoont slechts additieve effecten op de verlenging van de bloedingstijd geïnduceerd door oortransectie bij het konijn, wat aantoont dat gecombineerde remming van cyclo-oxygenase en ADP-effecten een aanzienlijk verbeterde antitrombotische werkzaamheid oplevert.
