 5.000mg/ml (14.91mM) Taking the 1 mL working solution as an example, add 50 μL of 100 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to clarify; then continue to add 500 μL of ddH2O to adjust the volume to 1 mL. The mixed solution should be used immediately for optimal results.

Biologische activiteit

Beschrijving 2-BP (2-Bromohexadecanoic acid) is een niet-metaboliseerbaar analoog van palmitaat en fungeert als een palmitoylation-remmer, die de activiteit van meerdere palmitoyl-acyl Transferase (PAT) remt ten opzichte van MyrGCK(NBD) en FarnCNRas(NBD) met identieke IC50's van ca. 4 µM. 2-Bromohexadecanoic acid kan de palmitoyltransferase-activiteit van alle DHHC (Asp-His-His-Cys) eiwitten direct en irreversibel remmen.

2-BP (2-Bromohexadecanoic acid) induces the interactions between GSDME-N and GSDME-C, and inhibits the GSDME-C palmitoylation, exhibiting chemotherapy induced pyroptosis.

  • Methode

    Cells are pre-seeded overnight until cell density reached ~60%. The culture medium is switched to fresh medium containing relevant drugs and incubated for a period of time as indicated. After treatment with 2-BP (2-Bromohexadecanoic acid) and TNFα+CHX, Hela cells are collected at the indicated time points and stained with FITC-Annexin V and PI, and subjected to Hoechst 33342 and PI double staining. The culture supernatants are collected to measure the percentage of LDH release.
